I can't say what the "right way" is for sure, but this is how I do it:
Cover letter - yes. I usually attach, but it doesn't matter, I think. Include some of their core values to show it's not a mass copy. Address CEO/partner/head of group you want.
I've been emailing this HR lady for the past 8 months at this mega-fund, one-a-month. Sometimes she replies, sometimes she doesn't. I've finally converted her into an ally and will have business lunch with her next week going over what the investment team is looking for.
The moral of the story is, in all likelihood, you won't get a response at first, but don't let that deter you. So send away and try again in a few weeks.
