I can't even comprehend your question but you should always say they are your first choice. And I don't think it's either a good or bad sign. It's a pretty standard question in an interview.
It's not a sign of any kind. You should always tell every bank you interview with that they are your first choice. It's somewhat childish, but people don't like to give offers only to be turned down.
