ArcGIS is pretty much the gold standard, most GIS data comes as shapefiles that can be loaded directly into ArcGIS so you overlay multiple data points on one map. Honestly, if just trying to find land 'for sale', that would be overkill. If it's 'for sale' just follow the advertisements or use loopnet, etc.
How do you recommend learning about ArcGIS?
Do most people use the free edition for Real Estate development? (It seems the free layers are pretty good)
Are there any layers/features you recommend? any layers for retail development?
You may want to look at ESRI Business Analyst (if you are a member of the CCIM Institute you get it as part of membership, with some limited features). It's easier to use than ArcGIS, and lets you do retail site analysis.
It's not for 'finding' sites, but really, I don't know why GIS would be used for that unless you want to organize a geo-database of parcels.
I feel like you have two different questions in there, the best GIS Software is probably ArcGIS, but it has nothing to do with land sales.
If you want to find land for sale, and need a map to do it, CoStar is going to be your best bet.
