Informational Interview Specifics

When having an informational interview should you lead with or end with what the main purpose of the call is?

If I am speaking to a Director at a boutique and I initially emailed him about internship opportunities should I introduce myself and be straight like are you guys looking for interns? Or should I ask him basic generic questions first then ask?

I've read up on questions to ask on this forum and online, but some of them seem like they lack depth. Could you guys recommend you questions you asked or would love to be asked?
