Did this person find the internship himself by cold-calling, etc? I am not saying it is necessarily so, but it sounds like this person may have gotten the internship through connections or someone helped him by making an introduction. The obvious giveaway is that the firm said they do not take interns on a regular basis.
If you are going to be in Tokyo anyways, then you certainly have nothing to lose. Asking for an informational interview is ok, but it might be better if you can reach out to as many people as you can at the firm before you arrive in Tokyo. Who did you initially contact at the firm? Was it HR? If possible, you should try to contact people who work in the front office (I am assuming this is the area you are interested in working in) Are you able to find profiles of people who work at this firm on linkedin?
